These seeds contain colchicine, 3-demethyl colchicine and colchicoside. Column chromatography studies using silica gel, polyamide and cellulose column fillers in certain volumes and at certain intervals (using the supercritical fluid extraction (SFE) method by performing 95:5 methanol-water extraction) has been done. The purity of the extracts was checked by both TLC and HPLC. As a result, the best extraction efficiency was obtained after 5 hours of agitation with 200 mL methanol-water solvent system. It was determined that there was 0.2-0.25% colchicine in the seeds. As a result of modified studies with SFE at 35 °C, 247 bar pressure, 1.5 mL/min flow rate and various ratios, a crude mixture containing around 0.2% colchicine was obtained.
